Ontogenic Study on Oxytocin Neurons in the Hypothalamus of the Rat by the Useof 5

Abstract
To determine the time of origin of oxytocin neurons in the hypothalamus, double immunohistochemical procedure was carried out with the use of hromodeoxyuridine (BrdU), anti-oxytocin and anti-BrdU serum. Groups of pregnant rats were injected with BrdU at the gestational day from 11 to 18, and were sacrificed after birth on the 60 days of age. Perfusion of the brain with paraformaldehyde solution was followed by serial coronal cryocutting and free floating staining. For the first staining, sections were incubated in anti-oxytocin serums, detected by avidin-biotin peroxidased complex and visualized by diaminobenzidine. Subsequently anti-BrdU serum was applied to the same seection and visualized by diaminobenzidine/nickel chloride. It yielded a distinct brown cytoplasm that is easily distinguishable from bluish-purple necleus in the paraventricular nucleus and supraoptic nucleus, BrdU-labeled neurons were located in the specimen which was injected BrdU on the gestational day from 13 to 16, and BrdU labeling index of oxytocin neurons was the highest at the gestational day 14 and 15. As a result, in the supraoptic nucleus and paraventricular nucleus, oxytocin neurons were formed mainly on the gestational day 14 and 15.
